    Conjugated Polymers Combined with a Molecular Beacon for Label-Free and Self-Signal-Amplifying DNA Microarrays

    A conjugated polymer (CP) and molecular-beacon-based solid-state DNA sensing system is developed to achieve sensitive, label-free detection. A novel conjugated poly(oxadiazole) derivative exhibiting amine and thiol functional groups ( POX-SH ) is developed for unique chemical and photochemical stability and convenient solid-state on-chip DNA synthesis. POX-SH is soluble in most nonpolar organic solvents and exhibits intense blue fluorescence. POX-SH is covalently immobilized onto a maleimido-functionalized glass slide by means of its thiol group. Molecular beacons having a fluorescent dye or quencher molecule as the fluorescence resonance energy transfer (FRET) acceptor are synthesized on the immobilized POX-SH layer through direct on-chip oligonucleotide synthesis using the amine side chain of POX-SH . Selective hybridization of the molecular beacon probes with the target DNA sequence opens up the molecular beacon probes and affects the FRET between POX-SH and the dye or quencher, producing a sensitive and label-free fluorescence sensory signal. Various molecular design parameters, such as the size of the stem and loop of the molecular beacon, the choice of dye, and the number of quencher molecules are systematically controlled, and their effects on the sensitivity and selectivity are investigated.Peer Reviewedhttp://deepblue.lib.umich.edu/bitstream/2027.42/64307/1/3317_ftp.pd

    Overexpressed Genes/ESTs and Characterization of Distinct Amplicons on 17823 in Breast Cancer Cells

    Abstract17823 is a frequent site of gene amplification in breast cancer. Several lines of evidence suggest the presence of multiple amplicons on 17823. To characterize distinct amplicons on 17823 and localize putative oncogenes, we screened genes and expressed sequence tags (ESTs) in existing physical and radiation hybrid maps for amplification and overexpression in breast cancer cell lines by semiquantitative duplex PCR, semiquantitative duplex RT-PCR, Southern blot, Northern blot analyses. We identified two distinct amplicons on 17823, one including TBX2 and another proximal region including RPS6KB1 (PS6K) and MUL. In addition to these previously reported overexpressed genes, we also identified amplification and overexpression of additional uncharacterized genes and ESTs, some of which suggest potential oncogenic activity. In conclusion, we have further defined two distinct regions of gene amplification and overexpression on 17823 with identification of new potential oncogene candidates. Based on the amplification and overexpression patterns of known and as of yet unrecognized genes on 17823, it is likely that some of these genes mapping to the discrete amplicons function as oncogenes and contribute to tumor progression in breast cancer cells

    Hybrid BCI Coupling EEG and EMG for Severe Motor Disabilities

    AbstractIn this paper, we are studying hybrid Brain-Computer Interfaces (BCI) coupling joystick data, electroencephalogram (EEG – electrical activity of the brain) and electromyogram (EMG – electrical activity of muscles) activities for severe motor disabilities. We are focusing our study on muscular activity as a control modality to interact with an application. We present our data processing and classification technique to detect right and left hand movements. EMG modality is well adapted for DMD patients, because less strength is needed to detect movements in contrast to conventional interfaces like joysticks. Across virtual reality tools, we believe that users will be more able to understand how to interact with such kind of interactive systems. This first part of our study report some very good results concerning the detection of hand movements, according to muscular channel, on healthy subjects

    A versatile microreactor platform featuring a chemical-resistant microvalve array for addressable multiplex syntheses and assays

    A versatile microreactor platform featuring a novel chemical-resistant microvalve array has been developed using combined silicon/polymer micromachining and a special polymer membrane transfer process. The basic valve unit in the array has a typical ‘transistor’ structure and a PDMS/parylene double-layer valve membrane. A robust multiplexing algorithm is also proposed for individual addressing of a large array using a minimal number of signal inputs. The in-channel microvalve is leakproof upon pneumatic actuation. In open status it introduces small impedance to the fluidic flow, and allows a significantly larger dynamic range of flow rates (∼ml min−1) compared with most of the microvalves reported. Equivalent electronic circuits were established by modeling the microvalves as PMOS transistors and the fluidic channels as simple resistors to provide theoretical prediction of the device fluidic behavior. The presented microvalve/reactor array showed excellent chemical compatibility in the tests with several typical aggressive chemicals including those seriously degrading PDMS-based microfluidic devices. Combined with the multiplexing strategy, this versatile array platform can find a variety of lab-on-a-chip applications such as addressable multiplex biochemical synthesis/assays, and is particularly suitable for those requiring tough chemicals, large flow rates and/or high-throughput parallel processing. As an example, the device performance was examined through the addressed synthesis of 30-mer DNA oligonucleotides followed by sequence validation using on-chip hybridization.     The phylogenetic affinities of the extinct glyptodonts

    Among the fossils of hitherto unknown mammals that Darwin collected in South America between 1832 and 1833 during the Beagle expedition [1] were examples of the large, heavily armored herbivores later known as glyptodonts. Ever since, glyptodonts have fascinated evolutionary biologists because of their remarkable skeletal adaptations and seemingly isolated phylogenetic position even within their natural group, the cingulate xenarthrans (armadillos and their allies [2]). In possessing a carapace comprised of fused osteoderms, the glyptodonts were clearly related to other cingulates, but their precise phylogenetic position as suggested by morphology remains unresolved [3,4]. To provide a molecular perspective on this issue, we designed sequence-capture baits using in silico reconstructed ancestral sequences and successfully assembled the complete mitochondrial genome of Doedicurus sp., one of the largest glyptodonts. Our phylogenetic reconstructions establish that glyptodonts are in fact deeply nested within the armadillo crown-group, representing a distinct subfamily (Glyptodontinae) within family Chlamyphoridae [5]. Molecular dating suggests that glyptodonts diverged no earlier than around 35 million years ago, in good agreement with their fossil record. Our results highlight the derived nature of the glyptodont morphotype, one aspect of which is a spectacular increase in body size until their extinction at the end of the last ice age.Facultad de Ciencias Naturales y Muse
